A. Personal Statement
My long-standing research interests lie in cancer stem cells(CSCs), the m olecular me chanisms of c ancer , and developing targeted cancer drugs . Our work is mainly centered on the identifi cation and functional ly characteriz ation of novel key molecule(s) involved in CSC survival and self-renewal, and the elucidation of the molecular mechanisms underlying tumor initiation, progression and metastasis. I have published 20 papers in professional peer-reviewed journals.

1. Clusterin in the survival and stemness of gastric cancer stem cell
CSCs have been implicated in cancer relapse and metastasis , which may be due to their capacity for uncontrolled growth, their resistance to chemo therapies and radiotherapies , and involvement in the Epithelial-Mesenchymal Transition. U nderstanding the weak spot of CSC s and targeting this vulnerability will provide more effective treatment options and/or long-term cure s for cancer patients . In 20 17 , I was the first to demonstrate that clusterin, a glycoprotein that acts in various biological phenomena, is critical for Gastric Cancer Stem Cell ( GCSC ) survival and self-renewal. Further studies indicate d that clusterin can be found in a complex with HSP90 and sustains GCSC survival by regulating Hsp90 function . ( Under review, International Journal of Cancer ).

2. Development of novel anti-cancer drug s
By screening a collection of drugs, we discovered that a FDA-approved drug for purpose other than cancer therapy, verteporfin (VP), blocks clusterin expression , decreases HSP90 client proteins in GCSC and reduces the growth rate of GCSC xenograft ed tumor s in animal models . VP is currently in clinical use for conditions other than cancer. Re tasking this drug will , therefore, be rapid and cost-saving as it is anticipated to overcome ( GCSC-mediated cancer metastasis, relapse and chemo-/radio-resistance ) that currently prevent effective and long-lasting cures for affected patients. 3. T he role and regulatory mechanism of Ras25 in gastric cancer
The function of Rab25 in cancer has long been under debate, and has gained increasing attention as a potential tumor therapy target over recent years. The clinical significance of Rab25, especially the prognostic significance, in gastric cancer had not been fully understood. I demonstrated that Rab25 has a pivotal role in tumor progression at the tissue protein level and revealed the prognostic significance of Rab25 expression in gastric cancer, suggesting Rab25 to be a potential therapy target of gastric cancer ( IJCEP 2017 ).
