A.? Personal Statement
The research of our team have always been focusing on the mechanism of tumor cell metastasis, including special extracellular matrix proteins involved in the regulation of tumor cell invasion and metastasis, molecular signaling pathways of tumor cell metastasis regulated by the specific cell surface receptors and the mechanism of tumor hematogenous metastasis promoted by platelets participating in the formation of tumor thrombus. Our research has been continuously supported by the National Natural Science Foundation in China (NSFC) with 3 regular grants and 1 key grants, 2 key grants of Natural Science Fund Project of Chongqing, 1 sub-grant of National 863 Project of China and 1 sub-grant of National Basic Research Program of China since 2004. Our team have published more than 60 papers in the peer-reviewed professional journals and I have mentored about 15 postgraduates (10 PhD and 5 MSc).

1.? TN-c involved in the regulation of pancreatic cancer cell invasion and metastasis
Interaction of cancer cells with stroma cells ? facilitates tumor progression by rebuilding the existing extracellular matrix (ECM) microenvironment. In the tumor, ? upregulation of Tenascin-C (Tn-C) expression potentially can ? alter tumor behavior. However, the molecular mechanisms ? by which tumor-stroma interactions affect the tumor microenvironment have not been well characterized. We found that fTn-C was ? increased in metastatic pancreatic cancer. The fTn-C expression ? correlated with MMP-2 activity. Tn-C ? deposition into the ECM requires participation of active ? MMP-2 and SF. The deposited Tn-C could promote pancreatic ? cancer progression.

2.? Roles of Fyn in pancreatic cancer metastasis
Compelling evidence indicates that the deregulation of several signaling molecules is associated with progression of pancreatic ? cancer. Of these signaling molecules, the non-receptor-tyrosine ? kinase, c-Src, is overexpressed in 70% of human pancreatic cancers ? and its activity is an important property of tumor progression. Fyn, a critical member of the SFK family mediates a variety of cellular processes, including T-cell receptor ? signaling, regulation of brain function and adhesion-mediated ? signaling. Our study showed that activation of Fyn ? promotes cell proliferation and metastasis in pancreatic cancer and Fyn requires ? HnRNPs and SR proteins to coordinate and regulate splicing of Bcl-x and ?integrin β 1 ?in pancreatic ? cancer cells, thus ? promoting the proliferation and metastasis of pancreatic cancer.

3.? Tumor-derived exosomal promoted pancreatic cancer metastasis
Tumor exocrine promoting tumor metastasis has become a new growth point in cancer research in recent years. We found that pancreatic cancer cells can promote the invasion and metastasis of pancreatic cancer by secreting specific tumor exosomes through a variety of pathways: non-coding RNA carried in the exosome acts both on tumor cells to promote cell phenotypic transition enhances tumor invasion and on endothelial cells to regulate cell adhesion and promote tumor metastasis.
